Off to the clubhouse on Friday (can't wait) unfortunately my brother who is on a later flight never managed to bag a 'G'.

Has anyone had much luck with using miles to upgrade at the airport?

If so would I be able to upgrade my brothers flight on his behalf?

It is certainly possible, there's been several success stories reported on here about G upgrades on the day! As you might expect it all depends on availability

If there is then yes you can use your miles to upgrade his flight

Isn't there also the potential issue of the class of the ticket you are trying to upgrade? The only time I tried this - a few years back, admittedly - I was told "yes that is possible. 50k miles for the leg, plus the cost of a small car to move your original PE ticket to an upgradeable fare class"
