In previous years I have booked MAN - ATL - RSW return online in PE. I have then looked for UC availability on the MAN- ATL segment, phoned and upgraded with points. The Delta segment has remained in Comfort +. Is this no longer possible?
I am reluctant to book separate segments due to possible delays and having to collect my suitcase in Atlanta on the way home.

That should still be possible. I did something similar in December.

Naturally it has to be ticketed on a VS 932 ticket though. If you book with Delta directly, this won't be possible.

We are currently in Naples and booked our flights last August. We booked PE and upgraded with points and have Delta Comfort and UC on our return later this month. So hopefully it is still an available option.

Does anyone know if this should be possible with a credit card upgrade voucher as well? i.e. a cash Premium multi-segment trip (looking at LHR > JFK > DEN, with the JFK > DEN part operated by Delta), upgrading the LHR > JFK segment to Upper with a credit card voucher? I wasn't sure if you can mix a reward segment with a revenue one (which would be the end result).

I’m fairly sure that’s doable. I’ve upgraded a single segment (LAX>SEA>LHR) with points where only the VS metal leg was upgraded. The voucher just negates the points so it should be fine. Be clear with the agent you only want to Upgrade the VS metal leg.
