Sure this is an easy question for you lot.

Used miles to purchase premium economy tickets using 37,500 per seat as at the time upper class seats were unavailable. Now the upper class ones have been released can I swap the premium economy tickets to the upper class ones for the additional 12,500 each and the additional taxes etc.

Yes, you can upgrade a reward fare using more miles. You may have to pay a bit extra for surcharges etc though.

There is no extra 'tax' i.e. APD to pay as the rate is the same in PE and UC.

However the extra fuel surcharge will kick in

Not sure exactly what it will cost you but in may it cost me £30 to change from U to G from MCO-LGW ( only upgraded the return leg )

Thanks all for your accurate and speedy responses. Each ticket now upgraded to Upper Class for the sum of 12,500 air miles and £30 each. y) :D
