For Delta it's as follows:

Economy (under 3000 miles) = 25tier points per one way flight
Economy (over 3000 miles) = 50tier points per one way flight

LHR>ATL = 50 TP's
ATL>MCO = 25 TP's
MCO>MSP = 25 TP's
MSP>LHR = 50 TP's

So should be 150 tier points in total.

They're Delta flights however, does the first leg as its all under DL33 count as one trip or two as it includes a change at ATL

Sorry yes, as it's all 1 flight number the whole LHR>MCO legs will just be 50 TP's

So total will be 125 tier points.

Unfortunately you've fallen foul of an annoying little trick widely used by the US airlines.

To all intents and purposes the LHR-ATL and ATL-MCO flights are completely separate, on different aircraft. Yet combining flight numbers means DL can market it as a "direct" flight from LHR-MCO, even though you have to change planes and clear customs & immigration in ATL.

When you get to ATL you could always ask to move to a different flight to MCO, if you're desperate for the additional TPs. The US airlines are much more relaxed about last minute changes than European airlines are, and that route runs more or less hourly so you'd have a decent chance if you wanted to.

My layover at ATL is only 90 minutes, so depending how long it take to clear immigration may leave everything tight. Unfortunately it during half term so prices were ridiculous and unless i wanted 12-20 hour layovers it was the best of a bad bunch,
