I have moved it to the travel section.
If you do a search on the phrase New York, you will get lots of threads that will help.
This one, or this one, for example - but there are a few others.
I will also leave this one running for any new tips.

For your first visit, the biggest tip I can give you is make sure you take comfortable shoes! You will do a lot of walking and your feet will hurt, but its also the best way to see NY. Its going to be really warm when you are there, so make sure you drink plenty of water as its a muggy/sticky heat.
Also, don't try and do too much, its very easy to try and cram a lot in while you are there, and you just end up rushing from one place to the next and not really enjoying it - select the absolute musts and then just see what else you get done. Central Park on a Sunday morning is a fav of ours, why not get a rowing boat on the lake? Something different but a lot of fun too.

As it's your first trip you're in for a treat! Would definitely aree with Attitude23 re the shoes. For a great view of Manhattan Island try the Rockerfeller Centre as from the top there you can take in the view inclusive of the Empire State Building when looking south and great views of Central Park when looking north. The night bus tour is also a must. For breakfast (or anytime) it has to be the Carnegie Deli - 854 Seventh Avenue, at 55th Street.
Most of all, enjoy its a great City!

Taking some jogging kit and go running in Central Park on a Sunday morning.
Empire State Building
Walk across Brooklyn Bridge
Shopping (wouldn't bother with Macy*s though)
Take a trip out the city down to Princeton if the hustle and bustle gets too much
Water taxi/Boat trip
Tour of UN
Metropolitan Museum of Art - currently Ancient Greek and Roman exhibition IIRC
See a show in the Theater District
Lunch in Little Italy

The panoramic revolving bar on the almost top (?72nd) floor of the Marriott Marquis hotel in Times Square is highly recommended. At night, they do a really good value buffet and drinks aren't a rip off either. Only problem is the seating area revolves but the buffet stations don't! Gets REALLY confusing after a few drinks - but very amusing watching everyone walking round trying to find their seats!
The restaurant on the floor above is really good too - but a bit pricey.
Enjoy.
