Twilight Check-in - upgrade enquiry then or AM?

Travelling from LGW on Tuesday and would like to use Twilight check in on Monday evening. There looks like there is a G available on my flight (dummy booking). Question is, do I ask at the Twilight CI or on the day of the flight for possible miles/money upgrade? Current fare code dependent of course and a/v of G!
