Currently planning our trip to Florida/New York in March and trying to determine the best NYC airport to fly into from Fort Myers - does anybody have thoughts on this. I am thinking that the immigration queues would not be so bad at La Guardia, but have never arrived there before and don't know what the traffic is like from there into the city. Definitely don't want to get caught up in the traffic jam from JFK into Manhattan in the early evening. Advice, please?

Haven't used Newark before but have done La Guardia for a domestic flight and think its a pretty good option in terms of distance to Manhattan. Also you won't have issues with immigration as la guardia is mainly if not all domestic flights. Hope this helps!

Firstly love flying in and out of RSW - such as civilised airport.

Used Jet Blue from JFK to RSW and Newark to RSW several times without too much stress regarding immigration and departure. Also used RSW into Boston for our connection with VS.

Are you using Virgin for a transatlantic flight as you don't mention where you are originating from?

Nice  like I said we have flown into both NYC airports and into Boston for our homebound VS flights.
In March '16, we did find that JFK was busy as far as checking in was concerned but that was after a horrendous journey up from Sarasota airport. We vowed never to use JFK again :-( unless we had stayed in Manhattan.

Other than Canada (which has US pre clearance) there are no international flights at LGA

LGA is closer to manhatten but there are no easy transit options to either JFK or EWR. It's also a mess at the moment due to major construction work

There won't be any immigration as you're on a domestic flight - you only clear immigration on your first port of call, ie, Miami.

I've flown RSW -> JFK, EWR and LGA. La Guardia is my favourite as it's the closest to Manhatten. I'm unsure if their improvement works are causing any traffic delays out of the airport at the moment. Newark took us the longest and we were stuck in traffic for an age. I never find JFK that bad. Yes, it's heavy traffic but it does move. As long as you allowed an hour you should be fine.

We've flown RSW to EWR several times. As said above, RSW is a very good airport, well staffed. No immigration issues as it's a domestic flight. I've not gone onward to Manhattan as we were staying with family is New Jersey. I prefer Jet Blue to United every time but YMMV.

Most delays we have had in getting to / from airports in NYC have been when in Manhattan rather than on the way there. The last couple of visits this has been largely eliminated as where we stayed has the Williamsburg Bridge at one end and the Holland Tunnel at the other.
