I can't see how you can do it. Change the cabin and it changes the whole fare.
I would suggest it is easiest just to make a phone call.

You could work out the miles required by doing two dummy bookings for two one way trips.
The taxes will be more than you will pay for the return.
Do a dummy Upper booking return and note the taxes.
Do a dummy PE return and note the taxes.
Add the taxes and divide by two and you MIGHT get an idea of what you have to pay on top of the miles calculated from the two one way bookings. It won't be accurate because taxes differ on the out and in leg.

Yes you can book mixed cabin reward flights online

1) From the main homepage enter your route and dates (you can but don't actually have to select 'miles' from the payment type list).

2) After clicking Find Flights on the next page click the miles button towards the top of the page under where it says 'Pricing'

(there was no Upper reward availability on the LHR>JFK leg on the date I randomly picked)

3) Any reward availability will show and then just select your cabins for each direction

It definitely isn't as easy as it used to be. There used to be a search "all cabins" option and a colour-coded grid would show specific cabin availability each day. Now you need to seach each cabin class separately to see at-a-glance availability.

The app will seach all cabin classes but only on one specific date, this is the best way if you're not flexible with dates.

I have had another go today and I realize where I was going wrong.
I was selecting the cabin right at the start, but you need to go right through to select out and return legs before choosing.
Not very intuitive but it works.

As mjb mentioned above, if you want to check multiple dates, you need to click the 'Flexible Dates' box when selecting dates and the calendar window pops up:

and then on the next screen, select you cabin, direct route and miles from the options at the top:
